dc.description.abstract | In mammals, the corpus luteum (CL) undergoes structural luteolysis, apparently by apoptosis of luteal cells. However, during the luteal phase, unknown factors seem to protect the CL. The present study was undertaken to identify the antiapoptotic factors in bovine CL. Cultured bovine mid luteal cells were exposed for 24 h to luteinizing hormone (LH), insulin-like growth factor 1 (IGF-1), prostaglandin (PG) E2 or PGF2α. All substances decreased caspase-3 mRNA expression in the cells compared with non-treated cells (P<0.05). Exposure to an adenylate cyclase inhibitor (2’,5’-dideoxyadenosine [DDA]) attenuated the inhibitory action of LH and IGF-1 on caspase-3 mRNA expression (P<0.05), while DDA did not affect caspase-3 mRNA expression in PGE2-treated cells. Additional exposure to a PKC inhibitor (straurosporin [SP]) had no effect on caspase-3 mRNA expression in PGF2α-treated cells. Furthermore, all four hormones stimulated progesterone (P4) production by cultured bovine luteal cells (P<0.05). DDA inhibited LH-, IGF-1- and PGE2-stimulated P4 production (P<0.05). In addition, SP inhibited PGF2α-stimulated P4 production (P<0.05). A specific P4 antagonist (onapristone [OP]) attenuated the inhibitory effects of LH and IGF-1 on caspase-3 mRNA expression (P<0.05), but did not affect PGE2- or PGF2α-treated cells. All four hormones prevented the cell death induced by tumor necrosis factor α (TNFα), interferon γ (IFNγ) and Fas ligand (Fas L) (P<0.05), and attenuated caspase-3 activity induced by TNFα and IFNγ (P<0.05). Furthermore, all four hormones increased the ratios of bcl 2 : bax mRNA and decreased Fas mRNA expression in the luteal cells (P<0.05). The overall results strongly suggest that LH, IGF-1, PGE2 and PGF2α act as anti-apoptotic factors in bovine CL by suppressing Fas and caspase-3 mRNA expressions as well as caspase-3 activity. | en_US |
